An enchantress disguised as an old beggar woman arrives at a castle during a ball and offers the host, a cruel and selfish prince name Luigi, adopted son of King Edmond III, an enchanted rose in return for shelter from a storm. When he refuses the offer, the enchantress reveals her true form and transforms Prince Luigi into a werewolf and his servants into ghosts before erasing the castle, himself, and his servants from the memories of their loved ones and everyone else in the town. She warns Luigi that the curse will be broken, only if he can learn to love another and have that person love him in return by the time the last petal of the rose falls, or else he will remain a werewolf for the rest of his life while his servants become ghost forever.
Several years later, in the Kingdom of Sarasaland, Princess Daisy, the book-loving daughter of King Richard and Queen Lillian, dreams of adventure. She constantly brushes off advances from Tatanga, a war purple alien as he is not the type of man Daisy wishes to marry. On Princess Peach, princess of the mushroom kingdom and Daisy's best friend, and Mario's, Peaches fiancé, way to Sarasaland to visit Daisy, they became lost in the forest and after narrowly escaping a pack of wolves they seeks refuge in Luigi's castle. However, the Luigi imprisons them when they "stole" a rose from his garden as a gift to Daisy. Even though Luigi recognizes his long-lost bother Mario, Mario didn't recognize him, since his memories with Luigi have been erase. When Mario and Peaches horse returns without them, Daisy ventures out in search of them, and finds them locked in the castle dungeon. Daisy asks Luigi, whom she recognize him as the werewolf to let her take their place; Luigi agrees and drags Mario and Peach away.
Belle befriends the castle's servants, including Sonic the Hedgehog ghost, Yoshi the dinosaur ghost, Sally Acorn the chipmunk ghost, Rosemary Prower the fox ghost, and her son, Tails Prower the two-tailed fox ghost. They invite her to a spectacular dinner. When she wanders into the forbidden west wing and finds the rose, Luigi scares her into the woods. Daisy is ambushed off-guard by the wolves from earlier, but Luigi the Werewolf rescues her, injuring himself in the process. As Daisy nurses his wounds, a spark develops between them. The ghost servants told Daisy that Luigi wasn't always royalty, and he used to be happy living a normal life with his biological parents and older twin brother, Mario. Although, after his parents passed away, Luigi and Mario were separated after being adopted from different parents. After being adopted by King Edmond III, a cruel king, Luigi became crowned prince of his kingdom. King Edmond then took the opportunity to harden his adopted son's heart through abuse to make him a vain and selfish ruler like himself. Of course, the castle servants secretly disapprove of the King's treatment towards Luigi, but they were too scared to do anything about it as they feared of any retaliation that King Edmond would inflict on them.
King Edmond's horrible treatment is what caused Luigi to develop an arrogant personality towards his own subjects during his young teen years. Daisy was shocked to find out that Luigi is Mario's long-lost brother, but never heard Mario mentioning having a brother. A few days later, Luigi the Werewolf shows Daisy a gift from the enchantress, a book that transports readers wherever they want. Daisy uses the book to visit her mother's childhood home in Paris, where she discovers a plague doctor's mask and realizes that her mother was forced to leave when her parents succumbed to the Bubonic plague, the one thing her mother couldn't tell her all those years.
In Sarasaland, Mario and Peach fail to convince the other villagers of the Werewolf and Daisy's imprisonment. Tatanga, seeing rescuing Daisy as an opportunity to win her hand in marriage, agrees to help Mario and Peach. When Mario and Peach learn of his ulterior motive and rejects him, Tatanga abandons them to be eaten by the wolves. Mario and Peach were rescued by King Richard and Queen Lillian, but when they told the townsfolk of Tatangas crime and is unable to provide solid evidence, Tantanga convinces them to send Mario, Peach, King Richard, and Queen Lillian to an insane asylum.
After sharing a romantic dance with Luigi, Daisy discovers her family and friend's predicament using a magic mirror. Luigi releases her to save them, giving her the mirror to remember him with. Arriving back to the town, Daisy reveals Luigi the Werewolf in the mirror much to the shock of the townsfolk, proving her family and friend's sanity. Realizing that Daisy loves Luigi, a jealous Tatanga claims she has been charmed by dark magic and has her thrown into the asylum carriage with her family and friends. He rallies the villagers to follow him to the castle to slay the Werewolf before he "curses" the whole village. Inside the asylum carriage, Daisy tells Mario that the Werewolf is his long-lost brother, even though he doesn't remember and told her mother that she already knows what happened to her grandparents and showed her the rose rattle she took from her magical visit to her old, abandoned home. Mario, Peach, King Richard, Queen Lillian, and Daisy escape, and rushes back to the castle.
During the battle, Tatanga abandons his minion companion, who then sides with the ghost servants to fend off the villagers. Meanwhile, he attacks Luigi in his tower, who is too depressed to fight back, but regains his spirit upon seeing Daisy return. He overwhelms Tatanga but spares his life at the last second before reuniting with Daisy and meeting his brother after being apart for a long time. However, Tatanga treacherously shoots the Werewolf from a bridge, which then collapses as the castle begins to crumble, leading Tatanga to fall to his death. Luigi then dies just as the last petal falls and the servants became ghost forever shortly after the ordeal.
As Daisy tearfully professes her love to the Werewolf, the enchantress appears and undoes the curse which repairs the crumbling castle, revives Luigi, and restores him and his servants to their normal flesh forms and to the villagers' memories once again. The movie ends with Luigi and Daisy hosting a ball at Sarasaland, where they dance happily.
