You receive the Holy Spirit by faith, similarly you receive further fillings of the Holy Spirit through faith. It is God's desire and command that his children stay filled with the Holy Spirit.

God does not take back the gifts once he gives them to you. You can use them at any time, although they are empowered supernaturally through the Holy Spirit, they are subject to you. You can start and stop at anytime with your free will.

"Fruits of the Spirit" is a biblical term that describe the visible attributes of a true Christian walk. It is visible physical manifestations of the work the Holy Spirit has done in your life. Its a sign of maturity as a believer.

There are barriers that can prevent you from receiving the Holy Spirit and even if you receive the Holy Spirit they can block or hinder his work. These are not barriers on His side as all things are possible for God. The only thing that is required is your consent and willingness to be worked on and change. A famous preacher once asked "Are you willing, to be made willing to allow God into your life?". A few common barriers are discussed here.
