When the exhibits at New York's Natural History Museum start behaving strangely, Larry and (Y/n) Daley and now the directors of nighttime operations must find out the cause. They learn that the Tablet, which magically brings Teddy Roosevelt, Jedediah, and the other exhibits to life at night, has started to decay. Larry and (Y/n) along with their son, daughter and museum friends, must travel to London's British Museum to learn how to prevent the Tablet's magic from disappearing.

Once the night guard at the Museum of Natural History, Larry Daley is now a successful purveyor of infomercial products. Soon after learning that his museum friends have been shipped to the Smithsonian Institution, Larry receives a distress call from Jedediah, who warns him that four of history's greatest villains have come to life and are planning to take over the Smithsonian first, then the world. Larry joins forces with his old friend (Y/n), along with other friends old and new to stop the quartet's nefarious plans.
