Red Dead Redemption tiene lugar en 1911, en la región (ficticia, como suele ser habitual en los juegos de Rockstar) de New Austin, sirviendo como una representación del momento en que el viejo Oeste y sus cowboys empezaron a desaparecer para dar paso al mundo del mañana. Bajo este marco empezamos a jugar con John Marston, un tipo de aspecto rudo (las tres cicatrices en su mejilla dan buena fe de ello) y un pasado aún más rudo.

Cinco años antes de los eventos de Red Dead Redemption, Marston era miembro de una banda de forajidos liderada por Dutch van der Linde, conocida como "la banda de Van der Linde". Tras llevar una vida de asaltos, atracos y asesinatos, Marston conoció en la banda a Abigail, de quien se enamoró y con quien tuvo un hijo, Jack Marston, y una hija que falleció. Durante un atraco a un banco, Marston fue herido de gravedad y dado por muerto, situación que aprovechó para huir de la banda junto a Abigail y el pequeño Jack. Y el primer objetivo en la lista es Bill Williamson, quien se esconde en un fuerte junto a un pequeño ejército.
