Izumi Midoriya as she is known now, spent those years like that, she was born thousands of years ago, but has lived longer than any human. Her life tragically ended when she was 15, but something forced her back to life, over, and over, and over...
She could never die, and her wounds were healed in an instant, but this was an ability she despised, for there was no one else like her, but with her power. She fought with armies of old, becoming a legendary warrior, her legacy was cemented.
But when you live as long as her, things are bound to change... In the 1600's, she was one of the many women who were accused of 'witchcraft', after being hung multiple times, and 'surviving'. This was all the evidence needed by the people, and they encased her in an iron coffin, dropping it to the bottom of the ocean... She escapes 200 years after, and her rage burns brightly, the 200 years she was trapped, and drowned over and over, she repaid kindly with 50 years enacting revenge on every human being she saw.

Black Blood:
When the affected person falls in love, their blood gradually turns black. When the affected person hasn't confessed their feelings, they will start to cough up the black blood. This disease is similar to the Hanahaki Disease.
