Gabriel Van Helsing, ein von der Kirche beauftragter Monsterjäger, wird nach Transsilvanien geschickt, um Graf Dracula zu vernichten. Dort trifft er auf Anna Valerious, die letzte Nachfahrin einer Adelsfamilie, die durch einen alten Fluch gebunden ist, solange Dracula lebt.
Während der Jagd begegnet Van Helsing auch Werwölfen, Frankensteins Monster und Draculas Bräuten. Im Laufe des Films wird enthüllt, dass Van Helsing eine mysteriöse Vergangenheit und eine Verbindung zu göttlichen Kräften hat. Das Finale führt zu einem epischen Kampf zwischen Gut und Böse in Draculas Schloss.
