Ayako grew up in a small town called Mishima, Shizuoka. She grew up in the typical Japanese middle-class society, her mother works as a paramedic nurse and her dad was a manga/illustration artist. When Ayako was around the age of 5, they discovered that she had an Ink Manipulation quirk, coming from her father, Taisei Sumitsuka, side of the family. Her mother, Kumiko Inoue, was home at the time of Ayako discovering her quirk. She was practicing writing with an ink set when she accidentally knocked the ink container over. Fearing that she would stain the carpet, Ayako held out her hand, stopping the ink from falling. In the process of discovering she could control ink, she played with it.

When her mother walked in to see her daughter playing with ink, she yelled, scaring Ayako. Ayako lost control of the ink, causing it to splash onto the carpet, apologizing to her mom. Kumiko told Ayako to go wash her hands while she called her father to tell him the news, she heard her daughter yell. Ayako, trying to wash her hands, began to notice that she couldn't wash the ink off her fingers, panicking, and called her mother for help. With Taisei on the phone, they tried to figure out why Ayako's fingers were stained black, finding out later that it was a side effect of her quirk. Ayako soon started elementary school, and other kids began to ask why her fingertips were stained black. Some students thought that she was evil, claiming that her fingers were stained from bad things she's done. Feeling upset, she asked her parents if she could start to wear gloves.

Her parents complied with her request, telling her that the kids don't know her quirk. Despite her struggles with elementary school and making friends, she was always seen smiling softly to herself when alone. As her dad is a jokester, telling jokes to cheer her up. He would often ask her to help him with his drawings. Because of this, she learned how to spend her time alone, often seen drawing by herself in school. While in her class one day, a student walked up to her asking what she was drawing. She hesitantly replied, the boy introduced himself as Daisuke Kuma, as the year went on they slowly became friends. As in sharing the same interest, Ayako thought he was interesting.
