important concl about MM kinetics:

          1. Characteristic of Km
          - reflects affinity of E for that S
          - numerically = [S] at which V = 1/2 Vmax
          - Does not vary with [E]
          - low Km, high affinity of E for that substrate
hight Km, low affinity of E for that substrate

bcs high concen. of S is needed to half-saturate the E

          2. Rship of V to [E]
ROR is directly proportional to [E] at all substrate concentration

          3. Order of rxn

MM EQN - describe how rxn velocity varies with substrate concentration

Assumption made in deriving MM rate EQN:
          1. [S] is much greater than [E]
          2. Rate of formation of ES complex is equal to rate of dissociation/breakdown of ES complex -> E+S and E+P
          3. ROR is measured as soon as E and S are mixed that is initial reaction volecities (V0) are used

how enzyme works?

          1. Energy changes occuring during formatiom rxn
          - Free energy of activation = barrier that separates reactant and product - difference in energy between reactant and intermediate occurs during formation of product
          - ROR - Low Ea, high Ea because molecule has sufficient energy to pass through intermediate state
          - Alternate rxn pathway - allows for lower Ea, accelerate ROR - enzyme does not change Ea of reactant, products and even EQ of the rxn

          2. Chemistry of active site
          - Transition state stabilization - Active site + substrate -> initiate conversion of substrate to transition state and accelerate ROR
          - Other mechanisms - active site provides catalytic groups that enhance transition state formation
