I think what really ignited this ship in the first place was all the "eye sex". In fact Misha has recently confirmed that in the script it literally says "Eye Fucking."
This part in their...relationship? Is impossible to go unnoticed. It's very obvious Dean and Castiel have many of these moments, and I honestly doubt this is just a friendship quirk.

Let's get to why Eye Sex means anything in the first place:
"Anytime you allow someone to look into your eyes you're allowing them to see how you feel at that moment. ... Their eyes will likely dart around the room as a means of "hiding". - the Art of CharmI think it's clear that's Dean's character likes to box up his emotions and play the tough guy, so when he allows Cas to see what he feels in that moment and let him read his emotions, it's kinda a big deal. He doesn't even let Sam do that? Right?
And also the obvious and plentiful sayings that if someone gazes into your eyes they like you or are attracted to you. Also Castiel is the only person Dean has these intimate eye moments with. I could talk about these two eye fucking all day, but I think you get the point.
