Harem: More girls that Joker could fuck, more awkward moments, harder to write, Kinda bland and cliché.
Single/Two: Less girls to fuck without making people pissed, More character and relationship development, Easier to write, Not as cluttered, less confusing, Less confusing, ect...
For harem you'll just have what is in the harem currently.
For Single/Two: Toga Himiko, Elena Allen or if you want another girl
