I would like to explain it, but I would rather not, so let's just dive right in!

First, we need a reminder of what the souls are and their traits. Below are the definitions (and yes, I copied them off Google, just because I can't think of better descriptions):
Red: Determination
[I] "The quality of being determined; the firmness of purpose."
Orange: Bravery
[I] "Courageous behaviour or character."
Yellow: justice
[I] "Behavior or treatment."
Green:Kindness
[I] "The quality of being friendly, generous or considerate."
Cyan: patience
[I] "The capacity to accept or tolerate delay, problems, or suffering without becoming annoyed or anxious."
Blue: integrity
[I] "The quality of being honest and having strong moral principles."
Purple: Preservance
[I] "Persistence in doing something despite difficulty or delay in achieving success."
Now that's out of the way, we can get to our first Boss Monster!

During Napstablook 's fight, he doesn't change your soul colour. This means that he is capable of all the traits, but due to his shy and timid characteristics, he hides it away.
In the pacifist route, you teach him how to love again. (You know which one I mean).
Love again? You may be confused. When Mettaton left Napstablook, he must have told himself how much everyone hates him;to have someone you love leave you like that.
However in the genocide route, you relentlessly keep attacking him, which was a dumb idea, because he is a ghost. But despite this, Napstablook dissappears anyways, as you reminded him that everyone hates him.

Just like Napstablook, Toriel (a.k.a Goat momma) doesn't change your soul colour. This, again, means that she possesses all the traits and it is completely evident withing the tut-TORIEL:
