The reason as to Why Kisame is Almost sure that the Puppy is Deidara is because of this.... It's only Kisame who can tell who you are from the point of Charka... Like let's say that if Kisame knows how your Charka Smell or if He/Samehada have had a Taste of you/your charka.. But it's Because of his sword Samehada is covered in scolding which absorbs his opponent's chakra and apparently only Kisame could use it... Also It's Only Kisame Himself knows about it.
