In the Rise of the Guardian movies the Tooth Fairy clearly says they're all born humans before and lived probably multiple human lives before they became Guardians. Sandman is the oldest Guardian in this story. But they are not some random space sprites that are immortal. Nope in fact Dreamworks Rise of the Guardians makes it clear below that there isn't another story credit to base the Rise of the Guardians off of otherwise it'd be listed on the screenshot screen below.
