Dean:
He will come over to you and the guy/girl and keep touching you. He'll put his arms around you, kiss your head and nuzzle your neck.
Sam:
He stays calm and watches for a while hoping to catch your eye and if you don't he'll hint that it's time to go.
Castiel:
He doesn't really understand jealousy but he recognises when someone's flirting with you so he'll come over and join you just so the guy/girl understands that you're taken.
