Based on the episode where Rachel and Puck are dating and in this one shot Rachel did know about Beth being Puck's and Quinn gave her blessing to Rachel to date Puck and Quinn is already off the cheerios.

Rachel was nervous, she normally wouldn't do something like this, as it wasn't her. She always wanted to save her voice and not yell or anything, but here she was standing outside the closed door of the gymnasium contemplating on whether or not to enter and whether or not she should join the team. She knew Quinn wasn't on the team anymore due to her pregnancy to Puck even though she was with him, Rachel still accepted and respected that Puck was going to be a dad, and every so often Quinn would ask Rachel to help them both especially with taking them to doctor's clinic and Rachel taking Quinn in and creating a nursery for Beth since her dads are never home. Rachel started pacing and thought about walking away from the close door or even running far away from there as possible, particularly when she heard their coach yelling.
Rachel stopped pacing as she made a decision, stood in front of the door, took a deep breath, placed her hand on the door, and entered the gymnasium for the cheerleading tryouts. She slowly made her way to where the other students trying out were and stood in front of the Cheerios and coach Sylvester. As she stood there, she heard the whispers from the other Cheerios asking why she Rachel Berry would want to join the Cheerios as she heard the whispers, she also heard
"Streisand, what the hell are you doing here in my Cheerio practice?" Rachel looked up and saw their coach Sue Sylvester waiting for her to answer. Rachel took a deep breath and said
"I'm here to try out for the team, I heard that a spot opened up since Quinn left because of her pregnancy." Rachel stood in front of everyone awkwardly thinking she made a big mistake with wanting to try out for the cheerleading team and thinking she wasn't even good enough to try out. Coach sue eyed her suspiciously as if questioning why she would join the team and then said
"Show me what you got." Rachel was shocked at those words and started her routine as the Cheerios sat on the bleachers and Sue sat in her seat and watched on. As they watched on, Santana was highly impressed as was Brittany because the girl they call names and insult had moves and routines for their cheerleading team and wouldn't even be surprised if she made captain, and most of all coach sue Sylvester was impressed by her routine, as she had never seen anyone that flexible or anyone that good at dancing or even someone do that complex of a routine. When Rachel had finished, she once again stood awkwardly and nervously in front of everyone in the gymnasium thinking her routine wasn't enough to make the team, as she also worried that maybe her boyfriend wouldn't like that she tried out for the team that used to bully her.
Rachel desperately wanted to make the team, as she tried her hardest with the routine and wanted something besides her boyfriend that's she just wanted for herself because everything was always picked out for her by her dads who were strict and wanted her to have the perfect life. Sue sat at her table, and eyed Rachel contemplating on whether she made the team or not and whether she would make the right decision. Sue lifted up her megaphone and said
"Young Barbra, pick up a uniform and put your hair in a high pony." At those words, Rachel was shocked, she walked up to the table, picked up the uniform, and as she said Sue said
"Everyone, including Barbra listen up." Rachel stopped walking, turned to face them and Sue continued
"Please welcome your new head Cheerio and captain. Rachel Berry." Rachel was shocked at those words and was shocked that she made captain after one try out, Sue knew that Rachel wouldn't let her down at all, and wouldn't let her team down either but she also hoped that she made the right decision. The other cheerios clapped while Brittany and Santana clapped the loudest, as they all walked out one by one congratulating Rachel on the way, with leaving her shocked and her walking to the change room and changing into the uniform and putting her hair in a high pony. Once she was changed, she placed her clothes in her bag and placed it in the locker room of the and then walked out. As she walked out and walking into the hallway of McKinley High everyone was looking at her shocked, she wasn't used to this much attention, and she didn't know whether she liked it or not
All the boys stared at her and stared at her long legs including Finn, but she wasn't interested in any of them especially Finn. she walked nervously up to her locker, thinking her boyfriend wouldn't like it or approve it. As she reached her locker, she was met with a wide smile and brown eyes that she fell in love with every time she saw him. She knew it was her boyfriend Puck. She stood there awkwardly and looked as Puck looked her up and down, fearing he wouldn't like the outfit. Rachel looked up at him and smiled nervously and said
"I made the cheerleading squad." Puck was pleased and proud of his girlfriend for making the team and said
"I can tell." Rachel then knew that he was pleased for her, and he then took her hand, twirled her around and said
"I love it." Rachel smiled in his arms and then said
"I'm also captain too." Puck was even more proud of her and then kissed her. Suffice to say he really did like the uniform, and was really pleased for her and proud of her, but most importantly he was pleased and proud that Rachel was his cheerleading girlfriend.
