Jensen puts a lot of thought into his acting and performances. That's a plain fact. Here are some brief analyzations (some may be quite obvious, but i'm listing them anyways) that lead me to believe that Dean is bisexual with a preference for women:

In Season 1's "Bugs", Sam and Dean investigate a new housing development together. Two of the employees assume that they are a couple and comment that the company accepts "homeowners of any race, religion, color, or..." with a smile "sexual orientation." The first time it happened, Dean states that they're brothers. The second time Dean smiles wide "Hm, right. I'm going to go talk to Larry, okay honey?" and then turns, smacking Sam's ass while walking away. I'm sure I am just looking a little too much into this, but I found it interesting that he wasn't exactly uncomfortable when saying "honey". He accepted the misunderstanding and played the part.
Season 2's "Playthings" episode presented another dilemma like that. The woman says that they "look the type." She continues, "So, a king sized bed?" Dean and Sam look a bit startled, and Sam is the one to actually correct her. Then Dean asks, "What did you mean by we 'looked the type'?" Could this be suggesting that Dean wasn't going to say anything at all, or possibly that Dean was wondering "Do I really look gay?" (By this episode it was the third time someone thought he was gay.)

In the fourth season, Dean and Sam are reading through comments on the books about their lives. Dean asks "What's a slash fan?" Sam leans back in his chair, "As in Sam/Dean... together." "Like, together-together?" "Yeah." "They do know we're brothers, right?"
Notice how Dean doesn't say, "They know we're straight, right?" Dean just focuses his disagreement on the fact that they're related, but doesn't address the actual gay aspect of it.

Garth asking if Dean is talking about the actual Purgatory, and Dean jokes "No, I'm talking about the one in Miami." Fun Fact: the Purgatory in Miami is a popular gay bar. Hm...

Sam teases Dean, "Are you strictly into dick now?" Alright, so he didn't say "Are you into dick now?" That would be Sam asking if Dean is gay. But he used the word "strictly", which turns everything around. "Strictly" means that Dean is into dick and vagina, and that Sam is asking if he's into only dick now. Do you see the bisexual hint?

There are certain men who just get him blushing, like Dr. Sexy. I mean, he was hardcore fangirling over him and knew every detail about Dr. Sexy like I would with Dean, so...
Dean was watching the show, and we can see him staring at a scene where Dr. Sexy is making out with a girl. Dean is interrupted by Sam, but later when he actually meets Dr. Sexy, there's a blatant attraction. "His face softens, his voice slips into a higher register, and he turns to Sam with a comically panicked expression on his face when he says, 'It's him; it's Dr. Sexy.' This reaction is similar to that of a teenager whose crush has just entered the room, which makes sense when one considers Dean's stunted emotional growth and the fact that crushes on fictional characters are safe." Loving a fictional character is considered "safe" because Dean (or any fangirl) will never have to face that fictional character, never have to go through anxiety if conversing with them, or face rejection.
