Trip would know how you act normally. Of course, he would, because he's your boyfriend and he pays close attention to any minute change in your personality. And so when you, a normally bubbly and happy and vocal person, are withdrawn, he'd know something is wrong.
He'd get very concerned. He'd notice it first during a brief break in the shifts; he'd either pass you in the hall or sit with you in the messhall, and he's just know something is up. He'd try to ask you if anything was wrong, but you'd just shrug it off and tell him you had to return to duty.
Trip would go out of his way to figure out what was eating at you. He'd ask your superior officer, ask your coworkers, and all your friends if anything had happened. Then, he'd go to Phlox to see if you'd gone in with a medical issue. Not that Phlox would be able to tell him if there was anything wrong, but since he (and virtually everyone) on the ship knew how serious you and Trip were, he would tell him whether or not you'd come in.
Reluctantly, he'd go back to his station for the remainder of his shift, but he'd be distracted. He'd replay your conversation and any interactions he had with you that day, trying to figure out what was bothering you. The second his shift was over, he'd go to find you. He'd find you in your quarters, alone. He wouldn't say anything when he got there, he'd just hold you, arms securely around your waist, and he'd kiss the top of your head.
After a few minutes of holding you in silence, he'd speak. "I know there's somethin' wrong, sweetheart. I know that pretty little head of yours is strugglin' with somethin'. I don't know what it is, but I am here for you, you know that, right? I'm always here for you. You can tell me anything and I'll do whatever I can to help."
You'd shrug and mumble under your breath, voice muffled by his shirt. He wouldn't press you or pressure you to tell him, he'd just repeat that you can tell him anything and that he loves you. When you were ready, you'd tell him what was on your mind, and he'd listen without interrupting. He'd rub your back, listening to you vent and tell him everything. And only when you were done talking would he say something. He'd offer you comfort, and support, and advice if you wanted it. And he truly would do whatever he could to help you feel better and help make the situation better, because he'd love you so damn much, and that's what loved ones do.
