Found this post on Facebook and I asked you all this question even though for some apparent reason out of the 2 people who responded one said Guts despite the fact he's not on this list.

That being said though Kaneki is in last place. Don't get me wrong Ken trying to hook up with a hot girl only to find out that said hot girl was actually a Ghoul that wanted to eat him and then get turned into a Ghoul in return is pretty brutal but it is nowhere near as brutal as the others especially since this is more of a "date gone wrong" type situation and not tragic loss. Sure he turned into a Ghoul but his circumstances are more of a walk in the park compared to the other three.
In third place I have Tanjiro. The world is to cruel for a kind hearted soul like him, all bro did was leave his home for awhile to help his village people then stopped to stay at someone's place after nightfall hits and was warned about demons only to return home the next morning and find his entire family all brutally massacred by a demon (that demon being Muzan of course) and then figuring out that Nezuko not only survived but turned into a demon. Thankfully Nezuko showed her human side proving that she still has humanity because had she not been able to prove it, Giyu would've most certainly killed her adding yet another layer of tragedy to Tanjiro which is heart breaking. That being said though the last two have suffered much worse than him.
In second place is Edward. Losing his mother is bad enough but it's even worse when both he and his brother Alphonse attempted to revive her by committing a taboo of Human Transmutation and as a result of their stupidity Ed lost his leg and Al lost his whole body and Ed had to sacrifice his arm just so Al's soul can resign in a suit of armor. That is bad that brutal doesn't even begin to describe it. Though they committed an unforgivable sin Ed and Al were driven by loss and tried to make things right which adds a whole another layer of tragedy.
But in the end Eren takes the number one spot of having the most brutal beginnings on this list. Not only did he lose his mother to man eating giant Titans, but he literally witnessed an entire genocide in his home town when the Colossal Titan broke down the walls of Shiganshina, Eren literally witnessed thousands of innocent lives die to Titans. This all adds another layer of tragedy because Eren eventually would go on to become a villain and final boss of his own story and commit complete mass genocide worldwide all because one country decided to fuck around and find out because there is so much loss one person can take and if you continue pushing them to a corner completely discriminating and segregating them they will become the very thing you accuse them of being and that's exactly what happened here, because Marley attacked Paradis at the beginning of the series causing Eren to lose his mother as well as make him witness some of the population being wiped out they have set the stage for Eren's journey from noble hero to genocidal villain that crushed 80% of the human population. As the saying goes: The more you fuck around, the more you're gonna find out. Yeah Eren definitely had the most brutal beginning no contest.
