Severus Snape held a long time grudge against Remus because Remus was a prefect but still didn't stop his friends from bullying him.
Snape became unhealthily curious about Remus's disappearances and since he was always on the Marauder's tails to find something horrible for them to get expelled, he suspected Remus of being a werewolf.
He was the only other student to find out about his lycanthropy as a result of a malicious prank played by Sirius on him by telling him about the passageway, hoping that he would be terrified or even get seriously hurt by what he found at the other end.
James was able to stop him and save his life in the nick of time, but Snape still uncovered Remus's secret, seeing him at the end of the passage.
