A lot of things abut John makes you laugh. One of the things that always gets a smile from you is his bluntness and bitchy streak. So when he tells Sherlock how much of a drama queen he is, you can't help but laugh.

When he accidently does something sassy you burst out laughing. Over time he's noticed this, and so now when he wants to make you laugh he purposely does something bitchy just to see your reaction.
