Of course it would be John. He's the first person Sherlock ever considered a friend. The two have been through so much together, it's only natural that Sherlock would choose him.
JOHN

Likewise, John chooses Sherlock. Sherlock is his closest friend, and trusts him beyond anything. Though Sherlock is terrified at first, and nerves almost make him say no, he steps up just as John knew he would.
MORIARTY

Jim doesn't trust anyone but himself, and he can't make himself his best man, so he decides not to have one. He is perfectly content to stand up at the top of the aisle alone and wait for you. The actual number of guests is very small too, so it does work out. But if you urge him to pick someone, he'll just chose one of his closest associates. It doesn't really matter to him as long as he gets to marry you.
LESTRADE

Greg has always liked John. He warmed to him the day the two met on a case involving a woman in pink. They usually stand to the side and give each other looks as Sherlock prances about solving the crime, and he's probably the closest friend Greg has because of how much time he spends with him on cases.
MYCROFT

Mycroft would definitely want Sherlock to be his best man. He loves his brother more than he would ever admit to his face, and so he'd want him next to him as he marries you. Sherlock is a little taken aback at first, but he wouldn't let his brother down.
